After a raft of halftime changes, Bournemouth's Rayan stretched the advantage eight minutes into the second period with Lucas Paquetá adding a fourth on the hour.
Brentford's Igor Thiago added a fifth from the penalty spot three minutes later after being fouled by Orlando Mosquera.
Danilo found the bottom corner from Lucas Paqueta's through ball to add a sixth before Carlos Harvey reduced the deficit late on for Panama.
Brazil complete their warmup games against Egypt in Cleveland on Saturday ahead of their opening World Cup game against Morocco on June 13.
